Is there a shortcut or any other way to stop playback once it started?
In Anki 2.1.54 for Windows you can pause the playback by clicking on the number “5”. I do not know about other versions. You can also click on the “More” button and then “Pause Audio”.

OMG! Thank you so much! I would never expect to see it in the “More” menu (I checked main application menu though)
